Auf einem Felsen über dem Neckartal gelegen ergänzt er das Gebäudeensemble um den Schillerhain, das unter anderem das Schiller-Nationalmuseum aus dem Jahr 1903, sowie das Deutsche Literaturarchiv, ein Bau aus den 1970er Jahren, umfasst.
Auf Grund seiner Lage bietet das neue Museum weite Ausblicke in die Umgebung, während in seinen introvertierten Ausstellungsräumen die reichen Bestände des Deutschen Literaturarchivs zur Literatur des 20. Jahrhunderts gezeigt werden, darunter das Maunuskript von Kafkas Roman Der Prozeß oder Döblins Roman Berlin Alexanderplatz.
Das LiMo wurde vom britischen Architektenbüro David Chipperfield Architects geplant und gewann den RIBA Stirling Prize 2007 des Royal Institute of British Architects und wurde beim Deutschen Architekturpreis 2007 ausgezeichnet. Projektarchitekt war Alexander Schwarz.
Der Bau verschmilzt Motive klassischer Architektur, wie die eines Peripteros, mit modernen Elementen und Materialien wie Holz, Muschelkalk, Sichtbeton und Werkstein.
